摘 要:脱离红细胞后的血红蛋白四聚体结构不稳定、氧亲和力强、半衰期短,还会引起一定的肾毒性。化学修饰作为一种新兴技术,能有效的改善上述不良特性。无基质猪血红蛋白(SFH)由新鲜猪血出发,经分离纯化制得。SC-mPEG大分子化pHb是延长其在循环系统中的停留时间、降低免疫原性、减少肾脏毒性的有效方法。幼犬的安全性实验表明产品无毒性,论证了其作为红细胞代用品的生物安全性。

Preparation and detection of the pig hemoglobin modified by mPEG
Abstract:Hemoglobin,when it is departed from red blood cell,has many disadvantages such as structure instability,high oxygen affmity,short half-life and kidney toxicity.As a burgeoning technology,chemical modification of hemoglobin has been applied to improve these poor characteristics of the hemoglobin.Stroma-free porcine hemoglobin is separated and purified from fresh pig blood by centrifugation.Increasing the total molecular weight of hemoglobin theough SC-mPEG coupling has been proved to be a good choice for prolonging vascular retention time,and it can reduce the immunogenicity and the kidney toxicity . Animal experiments with dogs showed their biological safety.
