摘  要:猪蓝耳病是猪繁殖与呼吸道综合征(Porcine reproductive and respiratory syndrome,PRRS)的俗称。根据猪繁殖与呼吸道综合征病毒(PRRSV)全基因组序列中的ORF7末端序列设计了一对特异性的引物P1/P2,扩增的片段为535bp,建立了猪繁殖与呼吸道综合征病毒的RT-PCR的快速诊断方法。特异性试验表明,这1对引物不能扩增其他常见的繁殖障碍相关病毒的RNA或DNA。试验结果表明本方法重复性好,敏感性强,可以检测到 10-4TCID50的病毒含量,提高了病毒的检出率和准确性,尤其是对于早期感染、潜伏期感染和持续性感染的诊断均具有重要的意义。本研究为更好地预防和控制猪繁殖与呼吸道综合征的发生和流行提供了技术支持。

RT-PCR detection for the PRRS
Abstract: Porcine reproductive and respiratory syndrome(PRRS) is also called “the blue ear disease of swine”. According to the sequence of ORF7 gene of the PRRSV, a pair of specific primer P1/P2 were designed, with the amplification fragment 535 bp, then RT-PCR was established to the rapid diagnosis method of PRRS. The specificity test indicated that the pair of primer could not amplify the RNA or DNA of other frequent correlated breeding obstacle virus. The result of test indicated that the method had high repeatability and strong sensitivity, capable of detecting the virus content of 10-4TCID50, increasing the detection rate and the accuracy. It also had a significant role especially for those primary infections, latent period’s infections and persistent infections. The research could provide the technical support for more efficient prevention and control the PRRS.
